Polarized infrared absorption of Na+/K+-ATPase studied by attenuated total reflection spectroscopy.
Na+/K+-ATPase can be isolated from the outer medulla of mammalian kidney in the form of flat membrane fragments containing the enzyme in a density of 10(3)-10(4) protein molecules per microm2 (Deguchi et al. (1977) J. Cell. Biol. 75, 619-634). In this paper we show that these membrane fragments can be bound to a germanium plate coated with a phospholipid bilayer. With this system infrared spect...
متن کاملIn situ monitoring of the DNA hybridization by attenuated total reflection surface-enhanced infrared absorption spectroscopy.
In situ monitoring of DNA hybridization kinetics is achieved via an attenuated total reflection surface-enhanced infrared absorption spectroscopy (ATR-SEIRAS) technique using a sandwich assay structure. The synergistic enhancement effect gives this ATR-SEIRAS-based detection strategy promise to be a convenient and unique platform for bioanalysis.

متن کاملAttenuated partial internal reflection infrared spectroscopy.
A new method for the spectroscopic study of absorbing films is proposed. In contrast to the well-established methods that take advantage of the attenuation of total internal reflection (ATR) to obtain spectra, we intentionally arrange the optics to permit partial internal reflection from the sampling prism face. Attenuated partial internal reflection (APR) spectroscopy is introduced through the...
متن کاملIncidence angle dependence of the enhancement factor in attenuated total reflection surface enhanced infrared absorption spectroscopy studied by numerical solution of the vectorial Maxwell equations.
The finite element method (FEM) was used to solve the time-harmonic Maxwell equations in a study of the effect of the incidence angle of infrared light on the surface enhancement caused by colloidal gold particles in attenuated total reflection surface enhanced infrared absorption spectroscopy (ATR-SEIRAS). The spectral enhancement factor was obtained from computations of absorbance from a thin...
